Evinacumab dramatically reduces cholesterol
Dramatic response to Evinacumab in a North Indian girl with homozygous familial hypercholesterolemia.
Most Relevant Research
We explored the effectiveness of Evinacumab, a monoclonal antibody recently approved for treating severe high cholesterol. Our focus was on a 16-year-old girl from North India diagnosed with homozygous familial hypercholesterolemia (HoFH) at the tender age of two. Despite her long-standing battle with high cholesterol, traditional treatments, including medications and lifestyle changes, failed to bring her cholesterol levels down to the recommended guidelines.

Genetic testing confirmed that she had a significant mutation affecting her LDL receptor, contributing heavily to her condition. Her LDL cholesterol levels hovered around 320 mg/dL even after trying the maximum doses of established medications. As her health deteriorated, necessitating aortic valve replacement surgery, we introduced Evinacumab into her treatment regimen. What we observed was truly remarkable; her LDL cholesterol dropped by over 76%, reaching a level of 82 mg/dL that has remained stable over the last four months.

This case highlights Evinacumab as a promising option for managing patients with HoFH, especially for those who have not responded to existing therapies. This was also the first instance of a child in India receiving Evinacumab for this condition, potentially setting a precedent for future treatments.

Cholesterol normalised
Most Useful Review
The result is positive. After two months of taking it alongside other supplements like rice, phytosterols, and omega, my cholesterol, which was critical, became normal. Omega and rice alone didn’t work. My husband also refused statins (which he had taken for three years) and his indicators are now normal.

Completely eliminated cramps
Most Useful Review
Great for leg cramps caused by statins! This truly works if you experience leg cramps while on a statin. It seems that statins tend to deplete the body's CoQ10. I began with 100 mg, but when that did not alleviate my leg cramps, my doctor advised increasing it to 200 mg, which utterly rid me of my leg cramps. Definitely worth the investment to stay on statins for cholesterol reduction.

YL-IPA08 shows promise against anxiety
The impact and mechanisms of YL-IPA08, a potent ligand for the translocator protein (18 kDa) on protection against LPS-induced depression and cognitive dysfunction in rodents.
Most Relevant Research
We investigated how a compound called YL-IPA08, which targets the translocator protein (TSPO), affects anxiety and cognitive function in a model of inflammation induced by lipopolysaccharide (LPS). This study incorporated a range of behavioral tests to assess anxiety-like behaviors and cognitive performance post-treatment. We also validated the use of YL-IPA08 with a TSPO antagonist to confirm that its effects were indeed connected to TSPO activation.

Our findings showed that administering YL-IPA08 significantly reduced anxiety and cognitive deficits associated with LPS treatment. Moreover, it was evident that YL-IPA08 notably affected the behavior of microglia cells in the brain. We documented a decrease in inflammatory responses and increased levels of beneficial factors necessary for mental health, indicating a robust protective effect against LPS-induced conditions.

Importantly, using a TGF-β1 inhibitor demonstrated that the anti-anxiety and cognitive benefits of YL-IPA08 are partly driven through this pathway. Thus, by targeting TSPO, YL-IPA08 offers a potential therapeutic avenue for managing anxiety and cognitive dysfunction caused by inflammation.

PTIC alleviates chest pain
Polymerized Type I Collagen Downregulates STAT-1 Phosphorylation Through Engagement with LAIR-1 in Circulating Monocytes, Avoiding Long COVID.
Most Relevant Research
We explored how polymerized type I collagen (PTIC) affects chest pain in adult patients suffering from COVID-19. In this study, symptomatic outpatients received PTIC, which was noted for its ability to lower inflammation and improve respiratory symptoms. One of the interesting observations was that these patients reported significant decreases in chest pain alongside improved overall conditions.

Our research indicated that PTIC impacts cells that play a crucial role in inflammation, specifically by interacting with a receptor called LAIR1. Through this interaction, PTIC was able to downregulate certain inflammatory markers, including those associated with chest pain and discomfort. The changes we recorded included a reduction in specific immune cells and their associated cytokines. Importantly, these findings suggest that PTIC may help alleviate chest pain as part of a broader improvement in symptoms post-infection.

While PTIC appears to have beneficial effects overall, it's essential to understand that its success in reducing chest pain was intertwined with other symptoms. The focus on chest pain improvement was part of a larger suite of results showing better oxygen saturation and diminished feelings of fatigue, cough, and dyspnea. Therefore, more research will be needed to fully isolate the specific effects of PTIC on chest pain alone.

No discomfort
Most Useful Review
I felt much better after taking this supplement. Previously, I experienced weird chest pains until I began using these regularly. My cardiologist had informed me that my heart tends to regurgitate blood a bit, causing minor discomfort. However, since I started taking these, I haven’t felt any discomfort whatsoever. I’m not sure if it’s just a placebo effect, but I intend to continue taking one daily for my heart health. It also aids in muscle recovery post-workout!

VEGFA, bFGF enhance heart recovery
Enhanced human adipose-derived stem cells with VEGFA and bFGF mRNA promote stable vascular regeneration and improve cardiac function following myocardial infarction.
Most Relevant Research
We aimed to uncover how the proteins VEGFA and bFGF can support heart recovery following a heart attack, and we did this by enhancing human adipose-derived stem cells (hADSCs) with modified mRNA. These proteins are known to play a role in improving blood flow and reducing cell death, both vital for effective healing.

By modifying the hADSCs to overexpress these proteins, we sought to improve their ability to support the heart during such a critical time. Importantly, our approach involved using a new technique that enables the hADSCs to produce these growth factors in a timed, efficient manner. This modification not only led to better survival of the transplanted cells but also promoted stable formation of new blood vessels at the site of injury.

We observed that the infused hADSCs improved overall heart function and electrical activity, suggesting a robust response to the proteins we introduced. The results indicate that the combination of these engineered stem cells and the proteins could substantially enhance recovery after a heart attack, paving the way for new treatments for heart diseases caused by ischemia.

MEDI0618 shows promise for migraines
Efficacy of MEDI0618, a pH-dependent monoclonal antibody targeting PAR2, in preclinical models of migraine.
Most Relevant Research
We observed the potential of MEDI0618, a targeted monoclonal antibody, in treating migraine by focusing on a receptor known as protease-activated receptor 2 (PAR2). This study analyzed how the antibody impacted migraine-like symptoms in various preclinical models, including both human and rodent systems.

Through a series of experiments, we found that MEDI0618 demonstrated a strong binding ability to human PAR2, effectively preventing increases in cell calcium levels triggered by certain enzymes. This suggests that MEDI0618 could play a key role in blocking pain mechanisms associated with migraine without affecting other related receptors.

The antibody was tested in several conditions simulating migraine pain, involving both chemical triggers and stress. Remarkably, MEDI0618 was effective in reducing headache-like pain across various models, including those not dependent on CGRP (calcitonin gene-related peptide), which is typically targeted in migraine therapies.

Although the findings are promising for the prevention of migraine attacks, it's important to note that MEDI0618 did not show effectiveness in alleviating post-traumatic headache pain from brain injury. Overall, this research highlights MEDI0618 as a potentially innovative treatment option for people suffering from migraines, especially those experiencing CGRP-independent episodes.

Immunoadsorption therapy's limited impact
Effect of immunoadsorption on clinical presentation and immune alterations in COVID-19-induced and/or aggravated ME/CFS.
Most Relevant Research
We aimed to understand how immunoadsorption therapy, which removes certain proteins from the blood, affects individuals with myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) following a COVID-19 infection.

Our study focused on 12 post-COVID patients experiencingME/CFS symptoms, heightened levels of specific autoantibodies, and neurological issues. We found that after receiving immunoadsorption therapy, there was a marked decrease in these troublesome autoantibodies associated with the autonomic nervous system.

Additionally, the therapy led to reduced levels of pro-inflammatory cytokines—substances involved in the body's immune response—which correlated with the presence of spike proteins in some patients. Despite these changes, we noted that, thirty days after therapy, while there were improvements in neuropsychological function and some physical strength, the self-reported symptoms and overall scores on ME/CFS questionnaires did not show significant improvement.

Therefore, while immunoadsorption therapy showed promise in altering certain immune markers, it did not lead to a substantial improvement in the patients' overall experience of ME/CFS.

Fermented pea protein improves sleep
Sleep-enhancing activity of fermented pea protein hydrolysate with enhanced GABA content by Lactobacillus brevis SYLB 0016 fermentation.
Most Relevant Research
We explored the potential of fermented pea protein hydrolysate, enriched with gamma-aminobutyric acid (GABA), to improve sleep. This intriguing study involved testing on ICR mice and Sprague Dawley rats, specifically looking at how this protein treatment could affect sleep duration and structure.

Our findings revealed that after fermentation by Lactobacillus brevis SYLB 0016, the GABA content increased significantly, which could be key in enhancing sleep quality. Notably, in tests where sleep was induced with pentobarbital, the hydrolysate extended sleep duration by an average of 25 minutes compared to the control group. Moreover, we found that the time spent in non-rapid eye movement (NREM) sleep increased, showing a boost in delta wave activity, which is important for restful sleep.

Further experiments indicated that the pea protein hydrolysate also benefited those suffering from caffeine-induced insomnia, effectively lengthening sleep periods at various dosages. Additionally, regular intake over three weeks improved GABA levels and receptor expression in the brain, which points towards a strong connection with sleep enhancement.

Overall, our research suggests that this hypoallergenic protein supplement holds promise for those struggling with sleep disorders. The findings support the idea that fermented pea protein could be a natural way to promote better sleep.

Exercise caution
Most Useful Review
Be careful! I took this in good faith but had an extraordinary reaction. I felt sluggish, nauseous, and unwell. After taking it at night, I woke up with heart palpitations. Reading the possible side effects helped me understand my reaction. CoQ10 users have reported slight stomach upset, headaches, feeling jittery, and experiencing mild insomnia. Other less frequent side effects include palpitations, anxiety, dizziness, and irritability. Just be careful with what you take. Thank you.
