When I gave my family member with hay fever regular niacin (500 mg) for the first time, I experienced stinging in my eyes and face, followed by severe dizziness and nausea. My neighbour also had stinging eyes. I even collapsed at the mall, prompting an ambulance ride to the hospital. Once there, I vomited significantly, which I rarely do, but afterwards, I felt revitalised and all my symptoms vanished. Surprisingly, this has worked quite effectively for them. Now, I’m giving a different formulation that doesn’t cause adverse reactions, and it seems to be fine.

We investigated how niacin affects nausea in mice suffering from a condition similar to pellagra. To mimic this condition, we fed the mice a low-niacin diet and administered isoniazid, a drug known to induce symptoms characteristic of niacin deficiency.

In our study, we observed that mice on a low-niacin diet displayed signs of nausea through pica behavior—a common way to assess nausea in rodents. Importantly, this behavior was not present in mice that continued to receive a normal diet.

After administering niacin as a treatment, we found that it effectively prevented the pica behavior and helped restore normal levels of certain metabolites affected by isoniazid. These findings suggest that niacin supplementation can significantly alleviate nausea-like symptoms associated with pellagra in our mouse model.

Overall, we’re excited that our newly established method for measuring pica could be an essential tool for understanding nausea associated with dietary deficiencies.

We conducted a study to understand how inositol, a naturally occurring sugar alcohol, influences anxiety levels in laying hens. Our focus was on the behavior of Lohmann LSL lite hens, which were fed two different diets—one rich and one deficient in protein—while receiving either pure inositol or high levels of phytase.

Throughout the experimental period from 19 to 59 weeks of age, we gathered data through multiple behavior assessments, which included observing their reactions to new objects and monitoring their overall health indicators like feather condition and mortality rates.

Interestingly, inositol appeared to help reduce fearfulness in these hens, which we observed through a decrease in the time it took for them to approach and peck at new objects. However, it's worth noting that while inositol also led to a drop in vent feather cover and was associated with a slight increase in mortality, its primary benefit was tied to its ability to lessen anxiety.

Phytase, on the other hand, did not yield similar effects on behavior or health. These findings point towards the potential of inositol as a useful dietary addition for improving the well-being of laying hens by reducing anxiety levels.

We aimed to understand the relationship between niacin intake and osteoarthritis (OA) by analyzing data from the National Health and Nutrition Examination Survey spanning from 1999 to 2018. With over 30,000 participants in the study and almost 2,000 diagnosed with osteoarthritis, we found some interesting results.

Our analysis used advanced statistical methods to explore how different levels of niacin intake affected the risk of developing OA. What we discovered was that higher niacin intake was consistently associated with a reduced risk of osteoarthritis. In fact, participants in the highest intake group had a remarkable 33% lower risk compared to those in the lowest intake group.

We also noticed that there’s a non-linear relationship; once we reach an inflection point in niacin intake, the way it impacts OA risk changes. The results were particularly striking in Non-Hispanic Black individuals and others in different racial groups.

Overall, this research suggests that by increasing the consumption of niacin-rich foods, we may be able to reduce the risk of developing osteoarthritis. However, further investigations are needed to confirm these findings and explore the potential of niacin as an effective dietary approach to OA prevention and treatment.

We investigated whether small extracellular vesicles (sEVs) derived from human palatine tonsil mesenchymal stem cells could boost myo-inositol levels in the context of menopause. This was particularly interesting given that menopausal women are prone to chronic inflammation due to physiological changes and related issues like metabolic syndrome.

To explore this, we treated mice with lipopolysaccharides (LPS) to simulate inflammation and assessed how the inclusion of T-MSC sEVs influenced myo-inositol production. After treatment, we measured levels of proteins involved in inositol synthesis in skeletal muscle and checked the serum for inositol levels.

Our findings are encouraging and suggest that T-MSC sEVs might help mitigate some effects of chronic inflammation linked to menopause. By supporting skeletal muscle mass and maintaining myo-inositol levels, these sEVs could be an effective strategy for improving metabolic health in menopausal women. This study sheds light on a potential new approach to tackle metabolic disturbances during this life stage.

Our study aimed to explore the impact of myo-inositol treatment on thyroid autoimmunity in women with autoimmune thyroiditis. We included groups of women who had been on myo-inositol for a while and those who hadn’t. For six months, we observed how vitamin D supplementation interacted with myo-inositol in affecting thyroid health and immune responses.

We found that among participants taking myo-inositol, the addition of vitamin D significantly improved insulin sensitivity and led to better outcomes regarding thyroid antibodies. Specifically, women who were already on myo-inositol showed a greater reduction in thyroid peroxidase and thyroglobulin antibodies when vitamin D was added to their regimen.

Interestingly, the effects of vitamin D were less noticeable in women who weren't on myo-inositol. These individuals did not exhibit the same improvements in thyroid function or antibodies. Overall, the findings suggest that myo-inositol might enhance the benefits of vitamin D, indicating a potential synergistic effect between these two agents in managing autoimmune thyroiditis.

We investigated the potential benefits of Myo-inositol, a naturally occurring sugar alcohol, on DU-145 prostate cancer cells. By treating these cancer cells with various doses of Myo-inositol, we determined its half-maximal inhibitory concentration (IC50) using a simple method that measures cell viability.

Our findings revealed that Myo-inositol significantly decreased the viability of DU-145 cells, showing an IC50 of 0.06 mg/ml. This suggests that Myo-inositol may effectively inhibit cancer cell growth.

Furthermore, we performed a detailed proteomic analysis using advanced techniques to explore how Myo-inositol changes protein expression. We found notable differences in protein levels between treated and untreated cells, particularly in areas linked to key cellular functions like cytoskeletal regulation, apoptosis (cell death), and stress responses. For instance, certain proteins tied to tumor suppression were upregulated in response to Myo-inositol treatment.

Overall, these observations highlight Myo-inositol's potential as a therapeutic option. However, further studies are necessary to unravel the underlying mechanisms and assess its effectiveness in combination with other treatments for managing prostate cancer, especially castration-resistant prostate cancer (CRPC).

Our research focused on understanding how inositol, specifically a molecule called 5-InsP7, plays a role in heart health by influencing levels of a substance called adiponectin. Adiponectin is important because it helps protect against cardiovascular issues and metabolic disorders.

We discovered that the enzyme inositol hexakisphosphate kinase 1 (IP6K1) produces 5-InsP7, which helps regulate how much adiponectin is available in the bloodstream. This regulation occurs through a process that ensures adiponectin stays stable instead of degrading within the cells.

When we decreased the levels of 5-InsP7—either by knocking out the gene responsible for producing it or through specific drug treatments—we noticed a significant increase in adiponectin levels in the plasma. This increase particularly favored the high molecular weight forms of adiponectin, which are particularly beneficial for heart protection.

The findings suggest that targeting 5-InsP7 could be a promising strategy for enhancing adiponectin levels, potentially offering a new avenue for protecting the heart against damage from conditions like ischemia-reperfusion injury, which occurs when blood flow returns to the heart after a period of blockage.

Overall, this study gives insight into how inositol treatment might help heart disease, emphasizing the importance of adiponectin in cardiovascular health.

We explored the potential of inositol hexakisphosphate (IP6) and its analogues as a treatment for calcium oxalate (CaOx) nephropathies, conditions often linked to kidney stones. Our study utilized a new machine learning approach that helped screen various chemical modifications of IP6 to find a particularly effective compound.

This innovative method led us to identify a divalent inositol phosphate that demonstrated remarkable activity. In our findings, this molecule completely inhibited the crystallization of CaOx crystals at very low concentrations. Additionally, it significantly reduced the interaction between crystals and kidney cells, preventing harmful changes in gene expression related to kidney damage.

We observed that, in a mouse model experiencing increased levels of oxalate, the IP6 analogue reduced crystal formation and subsequent kidney injury. With these promising results, we believe that these inositol analogues may offer a new therapeutic option for those suffering from calcium oxalate-related kidney issues.

We examined the link between dietary niacin intake and migraine occurrences by analyzing data from the National Health and Nutrition Examination Survey. This study focused on adults over 20 years old, looking specifically at their reported experiences with severe headaches and the amount of niacin they consume daily.

Among the 10,246 participants, we found that 20.1% reported having migraines. Those who had a higher intake of niacin showed a lower likelihood of experiencing migraines. For instance, compared to individuals with the lowest niacin intake, those in higher intake categories (Q2, Q3, and Q4) had significantly reduced odds of suffering from migraines. The results indicated that increasing dietary niacin had a favorable effect, suggesting an L-shaped curve relationship where the optimal intake appears to be around 21 mg per day.

This study highlighted a possible connection between niacin consumption and migraine prevention, indicating that moderate levels might play a protective role. However, it’s important to approach these findings cautiously, as they don’t establish cause and effect directly. Further research could illuminate how dietary choices might influence migraine frequency and severity.

We explored the potential of inositol, particularly Myo-inositol (MI), as a treatment option for depression, especially in individuals dealing with neurodegenerative disorders. The literature suggests that MI and its derivatives are vital in various cellular functions, including signal transmission and osmoregulation.

While our focus was primarily on neurodegenerative diseases like Alzheimer's, Parkinson's, and Huntington's, the interesting twist is how inositol may also help alleviate depressive symptoms. Research indicates that dietary supplementation with MI can have promising effects on mental health, suggesting a connection between inositol levels and mood enhancement.

However, despite the encouraging findings, much remains to be discovered. The relationship between inositol treatment and its effectiveness in treating depression is still being closely examined. Overall, it appears that while inositol shows potential, more comprehensive studies are needed to fully understand its benefits and mechanisms related specifically to depression.

We explored the effects of a synthetic heparin mimetic known as sulfated chiro-inositol (SCI) on blood clotting mechanisms. This innovative treatment aims to inhibit human factor XIa (FXIa), a key player in the clotting process, while minimizing bleeding risks typically associated with anticoagulants.

In our study, we synthesized SCI to engage FXIa specifically by targeting its heparin-binding sites. Through various blood coagulation assays and in vivo models, we observed that SCI effectively inhibited FXIa, leading to reduced clot formation without significant safety concerns. In particular, SCI demonstrated potent anticoagulant properties, matching the performance of existing treatments like enoxaparin but at a much lower dosage.

Importantly, our investigations revealed that SCI's effects could be reversed by common antidotes, which adds an important layer of safety. While we noted no major bleeding risks or toxicity in the studies, further research is needed to ensure SCI's efficacy across broader clinical scenarios. Overall, our findings highlight SCI as a promising candidate for safer anticoagulation therapies.

We explored the impact of inositol on cold stress in rapeseed, an essential oilseed crop affected by low temperatures. Notably, we found that inositol plays a significant role in enhancing cold tolerance in these plants by boosting calcium ion (Ca) influx.

Our study involved identifying various genes associated with inositol and analyzing their expression under cold conditions. Interestingly, inositol appeared to inhibit the expression of calcineurin B-like genes, which are involved in regulating calcium flow, suggesting a unique pathway through which inositol aids in cold resilience.

Additionally, when we overexpressed certain genes linked to calcium flux, we noted a stronger response to cold stress, further emphasizing the importance of the inositol-Ca pathway in facilitating cold tolerance in rapeseed. These findings not only deepen our understanding of how rapeseed copes with cold conditions but also present a potential strategy for enhancing crop resilience in the face of climate challenges.

We observed the effectiveness of Rebixiao granules, which include niacin, in treating patients with repeatedly attacking acute gouty arthritis. In the study, 90 patients were split into two groups. The first group received Rebixiao granule treatment, while the second group was given traditional anti-inflammatory tablets. Both groups also followed standard treatments like dietary changes and hydration.

One interesting aspect was the animal study that involved administering hypoxanthine and niacin to mice. This helped establish a model to see how these treatments affected uric acid levels in the blood. The results showed that Rebixiao granules were effective at reducing blood uric acid and had quicker results than just anti-inflammatory treatments alone. However, while niacin was part of the treatment, the study does not clarify its isolated impact on gout as it was combined with other ingredients.

Overall, we found that Rebixiao granules, possibly aided by niacin, were beneficial for gout patients without significant side effects. This indicates a promising option for managing gout symptoms and preventing future attacks.

Helps with hormones

Niacin, also known as vitamin B3, is a remarkable supplement. I take it during menopause, as it doesn't trigger hot flashes like regular vitamin B3. It aids those with hormone issues, helps with gout, and improves blood microcirculation. I wish you all good health.

We aimed to evaluate how myo-inositol supplementation affects women with gestational diabetes through a meta-analysis of randomized controlled trials. Our search included multiple databases, leading to the analysis of four studies involving 317 participants. The results showed that myo-inositol significantly reduced the need for insulin and improved insulin sensitivity. However, it did not influence birth weight, cesarean delivery rates, or NICU admissions. These findings suggest myo-inositol could be a helpful addition to managing gestational diabetes, but further research is needed.

We explored the potential of myo-inositol trispyrophosphate (ITPP) as a treatment for heart attacks and its effect on heart tissue injured by low oxygen levels. This research indicates ITPP modifies hemoglobin in red blood cells, enhancing the delivery of oxygen throughout the body.

Using an animal model for myocardial infarction, we observed that ITPP administration provided significant protection against heart failure and aided in recovery from left ventricular remodeling following the heart attack.

In our laboratory studies, heart muscle cells exposed to low oxygen levels reacted quickly to ITPP treatment, showing increased activity of a protein known as HIF-1. Interestingly, this overactivity was countered by ITPP when it was in the red blood cells, which suggests a strong connection between shear stress from blood flow and oxygen release facilitated by the modified hemoglobin.

Overall, the findings indicate that ITPP could be a game-changer in treating injuries to heart tissues related to low oxygen or ischemia. This approach may open new avenues for effectively managing damage caused by heart attacks.

We conducted a double-blinded, placebo-controlled clinical trial to explore how myo-inositol (MI) influences insulin resistance (IR) among obese individuals with non-alcoholic fatty liver disease (NAFLD). Our study involved 48 participants who were randomly assigned to receive either 4 grams of MI or a placebo daily for eight weeks.

Throughout this period, we measured various parameters such as weight, serum glucose levels, and insulin sensitivity indices before and after the intervention. Notably, we found that MI supplementation led to significant improvements in the expression levels of key genes related to the AMPK/PI3K/AKT signaling pathway, which plays a crucial role in regulating metabolism.

The results indicated noteworthy enhancements in insulin resistance for those who received MI, measured through multiple indices, reinforcing its potential benefits for liver health management. Thus, we observed that supplementing with myo-inositol not only bolstered the expression of important metabolic genes but also helped improve insulin resistance in our participants with NAFLD.
