We aimed to understand whether intranasal delivery of calcitriol, a form of vitamin D, can reduce inflammation in the nose and improve olfactory function after exposure to cigarette smoke. Our study used a mouse model, where the animals were subjected to cigarette smoke for nine months, simulating a chronic exposure similar to that seen in smokers.

Afterward, we treated these mice with calcitriol three times a week for four weeks. We observed that calcitriol significantly reduced inflammation and improved the overall condition of the sinonasal tissue. Specifically, our results showed an 18% reduction in inflammation severity compared to just a 5% reduction with smoke cessation alone.

In addition, we noted that calcitriol treatment reduced the influx of neutrophils—white blood cells that contribute to inflammation—and improved the mice's sense of smell, which had been impaired due to smoke exposure. These exciting findings suggest that local delivery of calcitriol could be a promising new approach for treating sinonasal inflammation and may offer benefits for individuals suffering from sinusitis.

However, the zinc levels in patients with eosinophilic polyps did not show a significant difference when compared to healthy controls.

While we saw a clear distinction in zinc levels between the neutrophilic group and the control group, the overall data suggests that zinc's role in sinusitis, especially for those with eosinophilic polyps, may not be as pronounced. As such, while zinc might contribute to inflammation in specific cases, its effectiveness as a standalone treatment for sinusitis remains uncertain.

I did take it for five days, but only after meals. On the fifth day, I took a capsule on an empty stomach and unfortunately, it resulted in vomiting. Nonetheless, this is a fantastic drug! I won’t take it again soon as I still have 95 capsules left. It’s now a must-have for colds.
