Addresses effects on ulcers
We explored the effects of a novel compound called salicyl-carnosine (SC), created from l-carnosine and acetylsalicylic acid (ASA), on ulcer prevention and its overall biological activity. Our research sought to assess whether SC could deliver the antioxidant benefits of l-carnosine while minimizing the gastrointestinal side effects often associated with ASA.

To gather our insights, we synthesized SC through the condensation of these two compounds. We employed various methods, including spectrophotometric and chemiluminescent assessments, to evaluate the antioxidant properties of SC. We also tested its antiplatelet activity using a light transmission-aggregometry method.

The results were promising; SC showed antioxidant capabilities similar to, or even better than, traditional l-carnosine. Additionally, we found SC's ability to inhibit platelet aggregation comparable to or exceeding that of ASA. Most notably, SC protected the gastric mucosa from ulcer formation and aided in healing existing lesions.

Overall, SC seems to retain the beneficial properties of both parent molecules, offering a dual advantage of antioxidant protection and reduced risk of gastric damage. This discovery opens exciting new avenues for developing drugs aimed at reducing inflammation and preventing blood clots without the traditional side effects of ASA.

Polaprezinc aids pressure ulcer healing
Oral treatment of pressure ulcers with polaprezinc (zinc L-carnosine complex): 8-week open-label trial.
Study shows potential benefits
We aimed to understand how l-carnosine, as part of polaprezinc, affects the healing of chronic pressure ulcers. In our open-label trial, we treated 14 patients suffering from various stages of pressure ulcers with 150 mg of polaprezinc daily for eight weeks.

Throughout the study, we monitored the severity of the ulcers weekly using the Pressure Ulcer Scale for Healing (PUSH) score. Remarkably, the PUSH score showed significant improvement, going from 8.1 at the start to -1.4 after eight weeks, indicating marked healing.

Of the 14 patients, 11 completely healed within the treatment period, and importantly, all patients completed the study without dropping out. We also noted increases in serum zinc levels, alongside decreases in serum copper levels and copper/zinc ratios, suggesting a well-tolerated treatment without serious complications.

These findings indicate that l-carnosine, when taken orally as part of polaprezinc, can potentially aid in healing pressure ulcers, providing a new avenue for treatment. Our results suggest l-carnosine’s safety and efficacy, inviting further exploration in larger studies to confirm these benefits.

We focused on the effects of polaprezinc (Pol), a compound that includes l-carnosine, and its ability to reduce inflammation in cells associated with neurodegenerative diseases. Through experiments involving LPS-induced BV-2 microglia, we observed that Pol successfully lowered the levels of nitric oxide (NO) and reactive oxygen species (ROS), which are often elevated during inflammatory responses.

Notably, Pol also suppressed the expression of key inflammatory markers, including iNOS, COX-2, IL-6, TNF-α, and IL-1β. These promising findings indicate that Pol could contribute beneficially to managing inflammation linked to ulcers.

Additionally, we investigated whether combining Pol with aspergillusidone G (Asp G) could further enhance its anti-inflammatory effects. What we found was quite remarkable: low doses of Asp G boosted Pol's ability to inhibit NO production from about 30% to an impressive 80%. Our analyses pointed toward the involvement of several molecular targets in this synergistic effect, further supporting Pol's potential role in addressing neuroinflammation.

Overall, while the study does suggest that l-carnosine, as part of Pol, plays a functional role concerning ulcers, it doesn’t isolate its specific contribution. Thus, more targeted research would be necessary to directly link l-carnosine's effectiveness to ulcer treatment.

We found that the hydrogel designed with l-carnosine and curcumin was not only biocompatible but also effective in the laboratory, promoting cell survival and reducing inflammation. Following tests on diabetic mice, we observed that this treatment significantly improved healing at wound sites, demonstrating the effectiveness of l-carnosine in this context.
