We investigated the role of melatonin in enhancing synaptic development in mouse models of autism. Through long-term oral administration over 28 days, melatonin improved social interactions and dendritic spine density in genetically modified mice.

Importantly, we found that melatonin's benefits are linked to the activation of a specific signaling pathway in the brain. However, when this pathway was blocked, the positive effects of melatonin on synaptic function were diminished. We explored melatonin's potential to impact neurodevelopment in autism by using stem cells from children with autism. In our study, we differentiated these cells into dopaminergic neurons and tested the effects of melatonin on their growth and function. We discovered that, without melatonin, these neurons showed reduced growth and mitochondrial dysfunction.

However, when melatonin was added, we observed significant improvements in neuron growth and mitochondrial health. Melatonin appears to enhance calcium signaling crucial for development. Excellent analogue of the pharmacy Melaxen. I gave melatonin to my son who struggles to fall asleep, especially when he's overexcited in the evenings. He is 6 years old and has autism. Children like him often have low melatonin levels, leading to sleep issues. Doctors often prescribe antidepressants that can harm not just the body but the brain. I sought a less harmful alternative and was recommended this melatonin. It calms my child effectively, allowing him to go to bed quietly without pills. I give him a quarter of a tablet, and he wakes up cheerful without headaches, which often accompany other medications. The price is very reasonable—240 tablets here were much cheaper than 24 in Russia. I highly recommend it!

As someone with Asperger's Syndrome, insomnia has plagued me for my entire life. I understand that those on the autism spectrum produce little or no melatonin naturally, hence the need for supplementation. I am very satisfied with NATROL brand melatonin and highly recommend it to mothers with children on the spectrum or adults in a similar situation.

We conducted a detailed look into sleep-related rhythmic movement disorder (SR-RMD) in children, involving data from 66 patients at a UK hospital.

Great product! I use it for my sister who has Down syndrome and autism, along with a herbal remedy prescribed medically, replacing strong sleep medications that caused her to drool. Gradually, we reduced the dose of her previous medication while increasing melatonin. Although melatonin alone couldn't resolve all her issues, today she enjoys peaceful sleep, wakes up in good humour and no longer drools. She's now more active and sociable, and I am very pleased with the product. I just need to order it a month in advance since it takes time to arrive in Brazil, but it's still cheaper than buying locally.

I sleep well. The melatonin from this manufacturer comes in various doses; 1 mg didn't work unless I took several. However, 5 mg was effective, and 3 mg was just right. It operates like a light sleep inducer, albeit overdosing can lead to lingering effects the next day. It has been beneficial for my children with autism and even my nearly 90-year-old mother who has dementia, as it helps them calm and sleep with just one tablet during times of excitement. The results are similar to other brands.

Melatonin and exercise comparably effective
We explored the effects of melatonin supplements, physical exercise, and their combination on sleep quality in children with autism spectrum disorder (ASD). Sixty-two children participated, assigned to groups receiving cycling, melatonin, both, or a placebo.

Results showed improvements in sleep efficiency and duration for all intervention groups, while the placebo did not yield significant changes. However, when comparing the groups, we found no notable differences between the effectiveness of melatonin and exercise, indicating both interventions could help alike.
