We conducted a detailed analysis to uncover the relationship between myo-inositol degradation by gut microbes and obesity. Through shotgun metagenomic sequencing, we examined a large group of 631 obese subjects alongside 374 normal-weight individuals. Our findings revealed that a particular cluster of gut bacteria, predominantly Megamonas, was more common among those with obesity.
Interestingly, we observed that the presence of Megamonas, along with genetic predispositions, appeared to synergistically increase the risk of obesity. We discovered that Megamonas rupellensis carries specific genes that help break down myo-inositol, a substance that can inhibit fatty acid absorption in our intestinal cells.
In experiments with mice, those given either M. rupellensis or a strain of E. coli that could also degrade myo-inositol showed higher lipid absorption in the intestines, leading to increased weight gain. Our research suggests that targeting the myo-inositol pathway may offer a new approach to obesity management.
