Oregano oil reduces lung cancer growth
Origanum majorana essential oil decreases lung tumor growth and metastasis in vitro and in vivo.

We investigated the potential of oregano essential oil, specifically Origanum majorana Essential Oil (OMEO), as a treatment for non-small cell lung cancer (NSCLC). In our pre-clinical study, we utilized two well-known human NSCLC cell lines, A549 and LNM35, to see how OMEO affected their growth and viability. To our satisfaction, we found that OMEO significantly reduced the viability of these cancer cells and inhibited the growth of their colonies in a concentration-dependent manner.

The oil worked partly by inducing a specific type of cell death, which is crucial for combatting cancer, and it reduced levels of a protein linked to cancer survival. We also observed that OMEO slowed down tumor growth in live models, including those implanted with A549 and LNM35 cells. Excitingly, it hindered the migration and invasion of these cancer cells and decreased the occurrence and growth of metastasis in the lymph nodes when tested in mice.

Remarkably, despite some three months of treatment with OMEO, the mice showed no adverse effects on critical body functions like blood or liver health. This suggests that OMEO not only offers a robust anticancer effect but also does so safely.

We isolated four specific compounds, known as depsides, from the medicinal plants Origanum dictamnus and Satureja pilosa to examine their effects on cancer cells. Using advanced techniques such as nuclear magnetic resonance and mass spectrometry, we identified these compounds and assessed their potency against human carbonic anhydrases, which are enzymes often associated with tumor growth.

In our tests, we found that these compounds preferentially inhibited tumor-associated carbonic anhydrases over the normal forms, suggesting a targeted approach in blocking cancer pathways. Furthermore, we employed X-ray crystallography to better understand how one of these compounds, salvianolic acid P, binds to the enzymes involved.

Importantly, we also observed significant cytotoxic effects on a particular type of breast cancer cell line known as MDA-MB-231. This finding indicates that these depsides could be promising candidates for further research in cancer treatment, opening new avenues for using natural compounds from oregano as therapeutic options.

We explored the therapeutic potential of oregano essential oil, specifically focusing on its ability to treat prostate cancer. Using a specially prepared form called oregano essential oil-based nanoemulsion (OENE), we aimed to see how it affected prostate cancer cells (PC3). Our methods included synthesizing and characterizing OENE, and then we conducted tests to determine its toxicity and efficacy.

Results revealed that OENE has an inhibitory concentration (IC) of 13.82 g/mL, meaning it can effectively reduce the growth of cancer cells. We observed pronounced changes in the cancer cells, such as shrinkage and decreased density. Additionally, OENE reduced the accumulation of harmful lipid droplets within these cells, pointing to its role in disrupting lipid metabolism.

On a molecular level, the treatment increased the expressions of pro-apoptotic factors like BAX and caspase 3, which help induce cell death. Meanwhile, the levels of the anti-apoptotic factor BCL2 dropped significantly. This combination of effects indicates that OENE can trigger apoptosis—the process of programmed cell death—in prostate cancer cells.

In summary, our findings suggest that oregano essential oil holds promise as a potential treatment for prostate cancer, paving the way for future in vivo studies.
