In our study, we utilized specialized nanoparticles to increase the effectiveness of quercetin, overcoming its usual limitations related to absorption and bioavailability.
Our research involved subjecting human chondrocytes to conditions that mimic OA. Afterward, we treated them with either free quercetin or our quercetin-loaded nanoparticles (Q-NP). We found that the Q-NP not only allowed for better cellular absorption but also selectively induced apoptosis, or programmed cell death, in senescent chondrocytes. This is significant because eliminating these unhealthy cells could pave the way for healthier joint repair and function.
In addition to promoting cell death in the right cells, Q-NP led to an increase in oxidative stress and reduced key markers of cellular aging. We also noted that the treatment halted the cell cycle in cells that needed to be eliminated. Overall, our findings suggest that enhancing quercetin delivery through nanoparticles holds great promise for improving osteoarthritis outcomes by actively removing harmful senescent cells from joint tissues.

We aimed to understand how N-acetyl glucosamine (GlcNAc), a compound known for its anti-inflammatory properties, affects the development and severity of collagen-induced arthritis (CIA). Our investigation involved applying a special multivalent GlcNAc-terminated glycoconjugate and assessing its influence through various methodologies.
The results were promising. We observed that GlcNAc treatment significantly reduced inflammation at the site of arthritis, notably decreasing the presence of immune cells that contribute to the disease. Furthermore, it appeared to hinder the activation of other immune cells, which are often involved in the production of harmful pro-inflammatory molecules.
Additionally, GlcNAc treatment not only delayed the onset of arthritis symptoms but also eased the severity of these symptoms in many cases. We found that about 18% to 31% of participants treated with GlcNAc completely avoided any arthritis symptoms. Overall, our findings support a new approach to managing arthritis by focusing on the glycobiological aspects of treatment, presenting GlcNAc as a potential therapeutic option for those suffering from this debilitating condition.

This is the first supplement that has truly improved my shoulder pain stemming from post-traumatic shoulder arthritis. We conducted a study to determine whether cat's claw, a medicinal plant from the Amazon, could effectively treat osteoarthritis of the knee. With 45 participants, we divided them into two groups—30 received freeze-dried Uncaria guianensis, while 15 were given a placebo. Over four weeks, we monitored their health through various assessments, including pain levels and any potential side effects.
Our findings indicated that cat's claw showed no harmful effects on participants' blood or liver function when compared to the placebo. Importantly, those taking cat's claw reported significant reductions in pain during activity and improvements in assessment scores within the first week. However, we did note that pain relief during rest or at night, as well as changes in knee circumference, were not significantly impacted in this short time frame.
In our lab tests, both U. guianensis and U. tomentosa demonstrated equivalent abilities to neutralize harmful free radicals and inhibit the production of TNFalpha, an inflammatory marker. Although cat's claw did reduce the release of another inflammatory substance, PGE2, this effect required higher concentrations than needed for TNFalpha inhibition. Overall, our study suggests that cat's claw could be an effective natural treatment option for managing knee osteoarthritis symptoms.

Notably, the prodrugs that included a specific amino acid called (L)-proline were particularly effective, even outperforming earlier versions.
Overall, our findings suggest that these novel compounds hold promise as new leads in the search for more effective treatments for osteoarthritis. They signify a positive step toward improving the management of this painful and degenerative joint disease.

However, a follow-up after three months of placebo indicated that while they felt better initially, their symptoms returned.
Importantly, EPO didn't change underlying disease progression, meaning it may not be a cure but can support symptom relief.

I purchased this supplement in despair due to my rheumatoid arthritis and hair loss, both undesired effects of this disease. To my amazement, after just 3-4 days of taking 2 capsules a day, I noticed a significant reduction in hair fall, which ceased completely after 10 days. My joint condition also improved significantly, despite not taking other joint supplements like MSM, hyaluronic acid, or curcumin. The inclusion of L-lysine (as HCl) in "Skin, Nails and Hair" was likely beneficial. After two months, my hair regained its shine, my nails no longer flaked, and my joints felt much better.
