Vitamin D derivatives enhance bone health
Synthetic Studies on Vitamin D Derivatives with Diverse but Selective Biological Activities.
Most Relevant Research
We looked into the potential effects of vitamin D3, particularly its derivatives, on osteoporosis. Recent research highlights how modifications to the A-ring of 1α,25-dihydroxyvitamin D can enhance its binding to the vitamin D receptor. This change not only boosts the vitamin's effectiveness but also helps it resist breakdown in the body, making it stay active for longer periods.

One standout example is a derivative known as AH-1, which demonstrated significant benefits for bone health in an osteoporosis model using ovariectomized rats. When given at a low dosage, AH-1 outperformed natural vitamin D, suggesting a promising path for improving osteoporosis treatment.

We also noted that while traditional vitamin D has its benefits, these newly developed analogs could lead to treatments that target osteoporosis more effectively, providing options without the side effects commonly associated with vitamin D therapy. This research emphasizes the importance of vitamin D derivatives as we seek better solutions for managing bone health.

A good dose of Vitamin D is crucial. I’m an endocrinologist and assert that it's a hormone rather than a vitamin. Testing for Vitamin D levels before supplementation is essential. The optimal level should be above 65 ng/ml, and dosages can be higher without adverse reactions. It's evident that Vitamin D enhances fertility in men and women, boosts mood, and helps prevent osteoporosis.

CYP11A1, Autism Symptoms, Vitamin D3
Placenta-specific CYP11A1 overexpression lead to autism-like symptom in offspring with altered steroid hormone biosynthesis in the placenta-brain axis and rescued by vitamin D intervention.
Most Relevant Research
We explored how overexpression of the CYP11A1 enzyme in the placenta can impact offspring and potentially lead to autism-like behaviors, particularly focusing on anxiety. Using a specialized murine model, we looked closely at how this genetic change affected social interactions and anxiety levels in the mice.

Our findings highlighted a significant increase in anxiety-related behaviors, which we measured through various tests like the open field and elevated plus maze assessments. Interestingly, we observed that supplementing with vitamin D3 led to noticeable improvements in these anxiety behaviors, suggesting a protective role for vitamin D3 against the adverse effects caused by altered hormone levels from the Cyp11a1 overexpression.

Additionally, we found that vitamin D3 treatment not only helped in behavioral aspects but also corrected underlying neuroinflammatory responses—specifically the activation of microglia in the brain. This suggests that addressing vitamin D levels during pregnancy could be a potential strategy to mitigate these changes and improve outcomes for offspring at risk of autism spectrum disorders.

Vitamin D3 ineffective for CFS
Effect of intermittent vitamin D3 on vascular function and symptoms in chronic fatigue syndrome--a randomised controlled trial.
Most Relevant Research
We conducted a thorough investigation into whether high-dose intermittent vitamin D3 could alleviate symptoms of chronic fatigue syndrome (CFS) and improve vascular health. This double-blind, randomized placebo-controlled trial involved patients meeting established criteria for CFS. Participants received either 100,000 units of vitamin D3 or a placebo every two months for six months.

Over the course of the study, we measured key outcomes such as arterial stiffness, blood pressure, cholesterol levels, and fatigue levels through various tests. Surprisingly, our findings showed that vitamin D3 treatment did not lead to any significant improvements in these areas. We observed only a slight increase in vitamin D levels in the treatment group, but this did not translate into better vascular health or reduced fatigue.

In conclusion, we honestly found that high-dose vitamin D3 did not offer any notable benefits for patients struggling with chronic fatigue syndrome. This study highlights the complexities of treating CFS and suggests that vitamin D3 may not be the solution some had hoped for.

Vitamin D3 shows no diabetes benefit
The effect of vitamin D supplementation on the incidence of type 2 diabetes in healthy older adults not at high risk for diabetes (FIND): a randomised controlled trial.
Most Relevant Research
We conducted a thorough investigation to understand how vitamin D3 supplementation influences the risk of developing type 2 diabetes in healthy older adults. Over the course of five years, we carried out a randomized, placebo-controlled trial involving 2,271 participants aged 60 and over. This diverse group of adults was free from cardiovascular disease and cancer, ensuring a focus on individuals not at high risk for diabetes.

Our study involved three groups: one receiving a placebo, another taking 1600 IU of vitamin D3 daily, and a third consuming 3200 IU daily. Remarkably, neither dosage of vitamin D3 showed a significant reduction in the risk of developing diabetes during our observations. Throughout the trial, we utilized national health registries to track the occurrence of diabetes, while a subgroup of participants underwent more detailed health assessments at various points.

Overall, despite the promising discussions around vitamin D and its health benefits, our findings indicate that vitamin D3 supplementation doesn't significantly lower diabetes risk in older adults who already have adequate vitamin D levels for maintaining bone health. This intriguing outcome adds valuable insight into the ongoing conversation about vitamin D's role in diabetes prevention.

Vitamin D3 impacts allergy severity
Correlation Between a Deficiency of D3 Levels and the Development of Allergic Rhinitis.
Most Relevant Research
We explored the relationship between vitamin D3 levels and allergic rhinitis (AR), a common condition characterized by sneezing, runny noses, and itchy eyes. In our research, we included two groups: thirty individuals with AR between the ages of 18 and 35 who had low levels of vitamin D3 and a control group of thirty people with sufficient vitamin D3 levels.

Our findings revealed a notable difference between the two groups. We observed that patients with AR had significantly lower levels of vitamin D3 compared to the control group. Additionally, there was a marked increase in immunoglobulin E (IgE) levels, which are often linked to allergic reactions, among those with AR.

Interestingly, our analysis indicated a clear inverse relationship between vitamin D3 levels and IgE levels. In fact, we found that changes in vitamin D3 levels accounted for about 58% of the variations in IgE among the participants. This suggests that increasing vitamin D3 might help reduce IgE levels and potentially lessen allergy symptoms.

Vitamin D3's role in OA prevention
1,25-Dihydroxyvitamin D Deficiency Accelerates Aging-related Osteoarthritis via Downregulation of Sirt1 in Mice.
Most Relevant Research
We explored the link between vitamin D3 and knee osteoarthritis (OA) by examining both young and older mice. Our findings revealed that a deficiency in vitamin D3, specifically the active form known as 1,25(OH)D, significantly sped up the development of age-related OA. This included issues such as cartilage damage and increased chondrocyte senescence.

When we supplemented with vitamin D3, we were pleased to see a reversal of these negative changes in OA phenotypes among the test mice. It not only improved cartilage health but also countered oxidative stress and cell aging associated with OA.

Our research highlights the important role of vitamin D3 in maintaining joint health and its potential to help prevent or slow down the onset of OA through mechanisms involving Sirt1, a gene linked to aging and cellular health. This suggests that ensuring sufficient levels of vitamin D3 could be a useful strategy for those at risk of developing arthritis.

Vitamin D3 modulates autoimmune response
Vitamin D3 attenuates autoimmune thyroiditis by regulating Th17/Treg cell differentiation via YAP/JAK1/STAT1 axis.
Most Relevant Research
We explored how Vitamin D3 impacts autoimmune thyroiditis, a specific autoimmune disorder. In this study, researchers established a mouse model of experimental autoimmune thyroiditis (EAT) to see how Vitamin D3 could influence this condition. After eight weeks of treatment, they assessed thyroid health and immune response through various methods, including staining tissues and measuring hormone levels.

Our observations revealed that administering Vitamin D3 helped reduce damage to the thyroid and decreased the presence of inflammatory cells within the thyroid. This treatment also led to lower levels of thyroid hormones and autoimmune antibodies in the blood, highlighting its positive effect on thyroid function. Additionally, Vitamin D3 appeared to promote the regulatory T cell (Treg) subset while decreasing the levels of Th17 cells, which are often involved in autoimmune processes.

Mechanistically, the study found that Vitamin D3 triggered specific pathways that influence Treg cell activity and survival. Notably, it activated a signaling pathway involving YAP and JAK/STAT, which are crucial for immune cell regulation. These findings suggest that Vitamin D3 could play a significant role in balancing immune responses in autoimmune thyroiditis, making it a promising candidate for further research and potential treatment strategies.

Vitamin D3 offers anti-cancer benefits
Liposomal encapsulation of cholecalciferol mitigates toxicity and delays tumor growth.
Most Relevant Research
We investigated the potential of vitamin D3, also known as cholecalciferol, in combating cancer while addressing concerns about its toxicity at higher doses. This study focused on enhancing the effectiveness of vitamin D3 through a clever method called liposomal encapsulation, which helps minimize side effects.

We prepared liposomal vitamin D3 (VD-LP) and conducted various tests to understand its capabilities. We found that this encapsulated form not only maintained high efficiency but also demonstrated improved stability. Our analyses showed that VD-LP had strong effects against cancer cells from colorectal, breast, and prostate cancers. Remarkably, it affected gene expression in immune cells, boosting elements that help fight infections and support the body's defenses.

Notably, VD-LP did a great job slowing tumor growth in mice and improved their survival rates without causing adverse effects like hypercalcemia, which is often a concern with regular vitamin D3 use. This evidence indicates that liposomal encapsulation of vitamin D3 could offer an effective cancer treatment strategy while minimizing unwanted side effects.

We believe this may pave the way for further research and clinical applications, showcasing vitamin D3 as a valuable addition to cancer therapy.

Vitamin D improves heart disease factors
The Effect of Vitamin D Deficiency Treatment on Lipid Profile and C-reactive Protein in Patients with Ischemic Heart Disease: Double-blind Randomized Clinical Trial.
Most Relevant Research
We conducted a double-blind, randomized clinical trial to see how treating vitamin D deficiency could influence heart health, particularly in patients with ischemic heart disease (IHD). In our study, 44 IHD patients aged 40 to 65 were treated with either vitamin D or a placebo to assess changes in their lipid profiles and C-reactive protein (CRP) levels.

Our findings revealed that participants who received vitamin D supplementation showed notable improvements. Specifically, we observed higher levels of high-density lipoprotein cholesterol (HDL-C)—often referred to as the "good" cholesterol—and lower triglyceride levels among those treated with vitamin D. Interestingly, the increase in HDL-C and improvement in triglyceride levels underscore vitamin D’s potential role in enhancing lipid profiles in patients struggling with heart disease.

We also noted that vitamin D treatment didn’t significantly impact CRP levels, which are typically used as a marker for inflammation and heart disease risk. This suggests that while vitamin D might help improve certain lipids, its effects on other markers of heart disease need further investigation.

Overall, our study highlights the potential benefits of vitamin D supplementation as a tool to manage heart disease risk factors, offering a promising option for healthcare professionals working with patients suffering from IHD.

Vitamin D3 improves depressive symptoms
Effects of vitamin D supplementation on depression and some selected pro-inflammatory biomarkers: a double-blind randomized clinical trial.
Most Relevant Research
We conducted a double-blind randomized clinical trial to explore the effects of vitamin D3 supplementation on depression. Our study involved 56 adults with mild to moderate depression, who were assigned to either a vitamin D group or a placebo group. Over the course of eight weeks, the vitamin D group received a high dose of 50,000 IU cholecalciferol every two weeks, while the control group received a placebo.

After the treatment period, we observed that the participants who received the vitamin D3 experienced a significant reduction in depression severity, as measured by the Beck Depression Inventory-II. Interestingly, this improvement in mood occurred independently of any changes in inflammatory biomarkers like IL-1β, IL-6, and hs-CRP, which we also assessed during the study.

This indicates that while vitamin D supplementation may positively impact depressive symptoms, its effects are not directly linked to the inflammatory markers we evaluated. Our findings suggest that vitamin D3 could be a helpful addition to depression treatment strategies, particularly in individuals with low vitamin D levels.

Vitamin D3 shows limited effect
Alterations in CD4 T Cell Cytokines Profile in Female Patients with Hashimoto's Thyroiditis Following Vitamin D Supplementation: A Double-blind, Randomized Clinical Trial.
Most Relevant Research
We conducted a double-blind, randomized clinical trial to investigate how vitamin D3 supplementation affects Hashimoto's thyroiditis (HT) in female patients. Over three months, participants received either 50,000 IU of vitamin D3 or a placebo. Our focus was on measuring changes in serum cytokine levels and gene expression in CD4+ T cells, which play a critical role in the immune response associated with autoimmune diseases like HT.

Following vitamin D3 supplementation, we observed a significant increase in serum levels of 25-hydroxyvitamin D, while serum calcium levels also rose compared to where they started. However, when checking for changes related to immune response, we noted that supplementation led to a decrease in interleukin-17 (IL-17) levels, but this change didn't differ significantly from those in the placebo group.

On the other hand, the transforming growth factor-beta (TGFβ) gene expression rose significantly with vitamin D3 treatment, yet once again, we didn't see a noticeable difference between the two groups. Importantly, there was no marked effect on other key immune markers, such as interferon-gamma and IL-4 levels. Although we did see some increase in T-bet and GATA3 transcription factors, the overall impact of vitamin D3 on Hashimoto's thyroiditis in our study suggests limited benefits.

This trial sheds light on the complex relationship between vitamin D supplementation and autoimmune conditions like Hashimoto's thyroiditis. While vitamin D3 shows some promise, further long-term studies are essential to truly understand its role and effectiveness in managing HT.

Vitamin D3 improves immune function
[Immunocorrecting properties and effect on the phosphorus-calcium metabolism of oxidevit (1 alpha-hydroxyvitamin D3)].
Most Relevant Research
We explored the impact of 1 alpha-hydroxyvitamin D3, commonly known as vitamin D3, on patients with chronic bronchitis, focusing on its potential benefits for the immune system and calcium-phosphorus metabolism. In this study, 45 patients received doses of either 2 or 4 micrograms of the vitamin over a period of 1 to 2 weeks.

Our findings revealed that the treatment led to observable changes in the metabolism of calcium and phosphorus. The alterations depended on the specific dose and duration of the treatment. Notably, the higher dose of 4 micrograms daily over two weeks was identified as excessive.

We also assessed various immune system components, such as total and rosette-forming cells, T-lymphocyte helpers and suppressors, and B lymphocytes. All these immune parameters showed significant normalization, regardless of the patients' initial levels. Elevated values tended to decrease, while those that were low increased back towards normal. It’s essential to note that despite these positive changes, the immune patterns remained stable throughout the treatment period.

Interestingly, we found a correlation between blood inorganic phosphorus levels and the dynamics of leukocytes, suggesting an interaction between immune responses and mineral metabolism. Overall, vitamin D3 treatment demonstrates a noteworthy effect on improving immune system function in chronic bronchitis patients, supporting its role as an adjunct therapy in managing this condition.

Calcitriol reduces age-related hypertension
Calcitriol reverses age-related hypertension via downregulating renal AP1/ATR pathway through regulating mitochondrial function.
Most Relevant Research
We explored the role of calcitriol, the active form of vitamin D, in addressing age-related hypertension. The study involved young and aged male mice, where we administered calcitriol daily for eight weeks. Our goal was to determine if this vitamin could effectively lower high blood pressure that often comes with aging.

Throughout the research, we measured blood pressure and examined various biological markers in the kidneys. What we discovered was quite promising. Aged mice exhibited increased blood pressure and impaired sodium excretion, both linked to changes in certain kidney pathways. After treatment with calcitriol, these mice showed significant reductions in blood pressure and improvements in sodium handling.

Mechanistically, calcitriol appeared to enhance mitochondrial function and alter the behavior of specific proteins related to blood pressure regulation. By promoting healthier kidney function and reducing harmful signaling pathways, calcitriol seems to offer a viable strategy for tackling hypertension as we age.

Overall, our findings suggest that calcitriol could be a valuable addition to our approaches in managing age-related hypertension, reaffirming the importance of vitamin D for our health as we get older.

Effect of Vitamin D Supplementation on Tibial Cartilage Volume and Knee Pain Among Patients With Symptomatic Knee Osteoarthritis: A Randomized Clinical Trial.
Most Relevant Research
We conducted a thorough investigation into the effects of vitamin D3 supplementation on knee pain and cartilage volume in people suffering from knee osteoarthritis, particularly those with low vitamin D levels. This study took place in Tasmania and Victoria, Australia, and was designed as a multicenter, randomized, double-blind, placebo-controlled clinical trial.

Over a span of two years, we divided participants into two groups: one received a monthly dose of oral vitamin D3 (50,000 IU), while the other group received a placebo. Our main focus was to evaluate any changes in tibial cartilage volume, measured through MRI, as well as the participants' knee pain levels as indicated by the Western Ontario and McMaster Universities Arthritis Index (WOMAC) pain score.

The results showed that, compared to the placebo group, those who received vitamin D3 experienced no significant differences in either tibial cartilage volume or reported knee pain over the two years. This indicates that vitamin D3 supplementation does not provide meaningful benefits for reducing tibial cartilage loss or improving knee pain in patients with symptomatic knee osteoarthritis.

Vitamin D3 alleviates MS symptoms
Immunoregulatory Effect of Calcitriol on Experimental Autoimmune Encephalomyelitis (EAE) Mice.
Most Relevant Research
We investigated the influence of calcitriol, the active form of vitamin D, on experimental autoimmune encephalomyelitis (EAE), an animal model akin to multiple sclerosis. Our study involved twenty-eight mice divided into four groups, allowing us to compare outcomes between those with and without EAE, as well as those receiving calcitriol treatment.

We found that, in mice with EAE, inflammation indicators such as interleukin-17 (IL-17) significantly increased, while transforming growth factor beta (TGF-β) levels decreased compared to healthy controls. Upon treating the EAE group with calcitriol, we observed a reversal of these changes, suggesting a substantial immunomodulatory effect, which eased symptoms typically associated with EAE.

Notably, we also noticed that calcitriol enhanced gene expression for interleukin-9 (IL-9) in both healthy and EAE mice. Overall, our findings support the notion that calcitriol holds promise as a potential treatment to alleviate the symptoms of multiple sclerosis through its anti-inflammatory properties.

Vitamin D analogs reduce acne effectively
Identification of a unique subset of 2-methylene-19-nor analogs of vitamin D with comedolytic activity in the rhino mouse.
Most Relevant Research
We explored the effects of vitamin D and its analogs on acne, primarily focusing on their ability to reduce the size of comedones (or acne lesions) in a rhino mouse model. This study evaluated the active form of vitamin D, known as 1,25-dihydroxyvitamin D(3), along with several modified analogs.

All analogs tested, including the natural hormone, increased the thickness of skin. However, only a particular group of analogs without a full side chain and 25-hydroxyl group were effective in actually reducing comedone size. Notably, this reduction in acne did not negatively impact serum calcium levels, which is a common concern with vitamin D treatments.

Interestingly, while all compounds featuring a side chain of 2 to 5 carbons worked similarly in terms of comedolytic activity, longer chains tended to increase calcemic risk. Moreover, our findings indicated that increasing the epidermal thickness required a lower dose compared to what was needed for comedolysis. This identifies a distinct set of vitamin D analogs that can help manage acne without the side effects of hypercalcemia.

Overall, we highlighted how certain vitamin D analogs can combat acne effectively while minimizing potential risks related to calcium levels.

Vitamin D3's role in anemia
Effect of vitamin D3-fortified fruit juice supplementation of 4000 IU daily on the recovery of iron status in childbearing-aged women with marginally low iron stores: Protocol for an 8-week, parallel group, double-blind randomized controlled trial.
Most Relevant Research
We conducted a study to see how vitamin D3 might help improve iron levels in women of childbearing age who had marginally low iron stores. Our approach involved a carefully designed 8-week randomized controlled trial, where 120 women aged between 19 and 40 were divided into two groups. One group received fruit juice fortified with 4000 IU of vitamin D3 daily, while the other had a placebo juice.

Throughout the study, we collected blood samples and monitored various health indicators every four weeks. We aimed to find out if the vitamin D3 supplementation could positively influence the iron status of the participants compared to the placebo.

Given the promising role of vitamin D in enhancing iron absorption, we hoped to uncover new insights into dietary strategies for managing iron deficiency. By observing the interaction over time between vitamin D levels and iron status, we aimed to provide valuable information on whether this fortification could serve as a practical alternative to traditional iron supplements.

As we gathered data, we explored the potential of vitamin D3 beyond its usual role, assessing if it could indeed help improve the anemia indicators in our target group.

Vitamin D3 alleviates angina symptoms
Vitamin D replacement therapy in patients with cardiac syndrome X.
Most Relevant Research
We aimed to understand how vitamin D3 treatment might help reduce angina and exercise-related ischemia in patients suffering from cardiac syndrome X and low vitamin D levels.

In our study, we treated patients with a course of vitamin D3 injections, delivered intramuscularly every other week for two months. We monitored the number of angina episodes per day as well as various measures of exercise capacity before and after the treatment.

By the end of the two-month protocol, we observed a striking increase in serum vitamin D levels to within a normal range. Most importantly, patients reported a significant reduction in angina frequency. We also noted improvements in exercise duration, work capacity, and a reduction in ST-segment depression during stress tests, which suggests a decrease in ischemia.

These findings support the idea that vitamin D3 replacement can dramatically improve the symptoms experienced by patients suffering from angina linked to cardiac syndrome X.

Vitamin D3 reduces breast cancer invasion
Mitigation of Breast Cancer Cells' Invasiveness via Down Regulation of ETV7, Hippo, and PI3K/mTOR Pathways by Vitamin D3 Gold-Nanoparticles.
Most Relevant Research
We set out to explore how Vitamin D3, when combined with gold nanoparticles (VD3-GNPs), could influence breast cancer's aggressiveness. Our research focused on breast cancer cell lines, specifically MCF-7 and MDA-MB-231, to see if this innovative treatment could significantly reduce cancer cell migration and invasion.

Through a series of methods including Western blots and migration/invasion assays, we observed that the VD3-GNP treatment led to an impressive reduction in cancer aggressiveness, with migration and invasion rates dropping by over 45%. This effect seems to come from the downregulation of key pathways associated with cancer growth and spread, namely the PI3K/AKT/mTOR pathway, as well as ETV7 and the Hippo pathway.

What's particularly surprising is the fact that we achieved these results using a relatively low dose of Vitamin D3—three orders of magnitude lower than doses used in past studies. This makes VD3-GNPs an appealing option for non-toxic and cost-effective treatments aimed at combating breast cancer's aggressive nature.

Erectile dysfunction unaffected by vitamin D
The effect of three years of vitamin D supplementation on erectile dysfunction: Results from the randomized placebo-controlled D-Health Trial.
Most Relevant Research
We aimed to understand how vitamin D3 supplementation affects erectile dysfunction among older men, drawing from data collected in the large D-Health Trial. This study involved Australian men aged 60 to 84, who were randomly assigned to receive either 60,000 IU of vitamin D or a placebo each month for up to five years.

After three years, blood tests showed significant differences in vitamin D levels between the two groups, with those receiving vitamin D averaging around 106 nmol/L compared to 76 nmol/L in the placebo group. However, despite these differences, we observed no significant change in erectile dysfunction prevalence. The rates were nearly identical, with about 58.8% in the vitamin D group and 59.0% in the placebo group, indicating that vitamin D supplementation did not improve or prevent erectile dysfunction.

In our exploration, we found no evidence that vitamin D had any beneficial effect on erectile dysfunction, regardless of the participants' characteristics. Thus, it appears that supplementing older men with vitamin D might not lead to any improvements in this area.

Vitamin D3 may alleviate fibromyalgia
Efficacy of Vitamin D Supplementation in the Improvement of Clinical Status in Patients Diagnosed with Fibromyalgia Syndrome: A Systematic Review.
Most Relevant Research
We conducted a thorough review of the existing literature to understand how vitamin D3 supplementation affects those suffering from fibromyalgia. This condition is often marked by widespread pain and fatigue, and we wanted to see if vitamin D3 could help alleviate these troubling symptoms.

In our search, we scoured various databases for studies published between January 2011 and May 2021. This included randomized controlled trials, observational studies, and other well-documented research focused explicitly on fibromyalgia. Out of the 2,651 studies retrieved, 12 met our strict criteria for inclusion, of which 11 were of high quality, showing minimal bias.

The results were promising, as 11 of those high-quality studies indicated a significant positive impact on the clinical status of fibromyalgia patients who received vitamin D3. This suggests that vitamin D3 might play an important role in managing fibromyalgia symptoms.

While our findings point towards the benefits of vitamin D3 supplementation, they also highlight the need for further research. As we continue to explore effective treatments for fibromyalgia, vitamin D3 stands out as a potentially beneficial option worth considering for patients seeking relief.

Vitamin D3 aids gastritis relief
[Vitamin D3 alleviates the gastritis that associated with Helicobacter pylori infection in mice with hypercholesterolemia by enhancing the activity of vitamin D receptors in the liver tissue and blocking the signaling pathway of JAK/STAT3].
Most Relevant Research
This study focused on how vitamin D3 (VD3) might help alleviate gastritis, particularly in the context of Helicobacter pylori (Hp) infection and high cholesterol levels. We established mouse models that reflected these conditions and then treated them with VD3 for eight weeks. After treatment, we observed significant positive changes in the liver and stomach tissues of the mice.

Notably, we found that the levels of vitamin D receptors (VDR) and a related protein called Insig-2 increased in the liver of the mice that had high cholesterol and were also infected with Hp. This indicates that VD3 enhances the activity of these important receptors, which might contribute to its gastroprotective effects. Additionally, we saw a reduction in serum gastrin levels, which can be important in regulating stomach function.

We also noted decreased expression of JAK and STAT3 proteins, which are part of a cellular signaling pathway known to be involved in inflammation. Delving deeper, the treatment with VD3 led to lower levels of COX2—a protein associated with inflammation—in the gastric tissue. Furthermore, serum cholesterol levels decreased, and we observed a reduction in inflammatory markers like IL-8.

Ultimately, our findings suggest that VD3 may offer a beneficial approach to managing gastritis by modulating inflammation and improving liver function, especially in those with high cholesterol and Hp infection. We can cautiously conclude that VD3 has promising potential in this area, although further research could solidify these findings.

Vitamins D3 and B2 inhibit gout
The inhibitory kinetics and mechanism of dietary vitamins D3 and B2 on xanthine oxidase.
Most Relevant Research
We investigated how dietary vitamins D3 and B2 can influence gout by inhibiting an enzyme called xanthine oxidase (XO), which plays a significant role in gout development. Through various analytical techniques, we found that vitamin D3 works by fitting into the active site of XO, effectively blocking it from interacting with its substrate, xanthine. This action is due to D3’s ability to form bonds with the enzyme, which leads to a more compact structure that restrains xanthine binding.

On the other hand, vitamin B2 showed a mixed-type inhibition. It binds near the active site, impacting the enzyme's flexibility and structure. This alteration can fold over sections that block substrate access, thereby reducing the enzyme's activity. Remarkably, we noted a synergistic effect when both vitamins were present, heightening their ability to inhibit XO within specific concentration ranges.

Our findings shed light on how vitamins D3 and B2 could be valuable in the dietary management of gout, especially regarding their potential roles as supplementary treatments. Overall, we believe these insights might encourage further exploration into the use of vitamins in supporting gout therapies.

Vitamin D3 lowers cholesterol levels
Vitamin D Supplementation Reduces Hyperlipidemia and Improves Bone Mass in Pediatric Obesity.
Most Relevant Research
We investigated how vitamin D3 supplements impact cholesterol levels in pediatric patients suffering from obesity. The study involved 723 children over two years, with 283 receiving vitamin D3 and 440 on a placebo.

After closely examining the children’s cholesterol, triglycerides, and glucose levels, we found a strong relationship between these factors and their vitamin D3 levels. Following the supplementation, the group taking vitamin D3 experienced some notable improvements.

Specifically, we saw significant decreases in total cholesterol, HDL cholesterol, and triglycerides in the vitamin D3 group when compared to those who received the placebo. This indicates that vitamin D3 could play a beneficial role in managing high cholesterol and improving overall metabolic health in children coping with obesity.

Our findings suggest that vitamin D3 is a valuable addition to strategies aimed at improving lipid levels and encouraging healthy lifestyles in young patients struggling with excessive weight. Overall, supplementing with vitamin D3 appears to offer an effective approach to tackling high cholesterol in this at-risk population.

Vitamin D3 inhibits lung cancer growth
Inhibition of lung cancer by vitamin D depends on downregulation of histidine-rich calcium-binding protein.
Most Relevant Research
We conducted a study to understand how vitamin D3 influences lung cancer, particularly focusing on a protein known as histidine-rich calcium-binding protein (HRC). Our research explored whether vitamin D3 could help lower HRC levels, given its crucial role in calcium balance, which is significant in cancer development.

We measured the expression of HRC and the vitamin D receptor (VDR) in lung cancer tissues compared to normal and adjacent non-cancerous tissues. Using various lab techniques, we assessed how vitamin D3 affects lung cancer cells, including their growth, spread, and ability to undergo programmed cell death, known as apoptosis.

Our findings revealed that vitamin D3 does inhibit the expression of HRC, which in turn decreased the migration and proliferation of lung cancer cells. Additionally, we noted an increase in the number of cells undergoing apoptosis in response to vitamin D3 treatment. In our animal model, mice treated with a vitamin D3 formulation exhibited smaller tumors and fewer cancerous cells than those treated with a combination of vitamin D3 and HRC.

Overall, our study indicates that vitamin D3 plays a protective role against lung cancer by downregulating HRC, thus slowing tumor growth and promoting cancer cell death.

Improved Healing of Diabetic Foot Ulcers After High-dose Vitamin D: A Randomized Double-blinded Clinical Trial.
Most Relevant Research
We set out to explore how different doses of vitamin D3, specifically Cholecalciferol, impact the healing of chronic diabetic foot ulcers. Our study included patients with diabetes who had one or more foot ulcers present for longer than six weeks. We assigned participants at random to receive either a high dose of 170 micrograms or a low dose of 20 micrograms daily. Throughout the experiment, both patients and assessors were kept unaware of who was receiving which treatment to ensure unbiased results.

Over our 48-week follow-up period, we observed that the high-dose vitamin D3 treatment yielded significantly better healing outcomes. Among the patients receiving the high dose, 70% of their ulcers healed, while only 35% of those on the low dose showed healing. The median ulcer reduction in the high-dose group was impressive, showing complete healing in many cases, while those on the low dose saw a more modest reduction.

In conclusion, our findings suggest that high-dose vitamin D3 can play a crucial role in promoting the healing of chronic diabetic foot ulcers. By effectively boosting healing rates, this treatment could provide an important avenue for improving health outcomes in patients suffering from these persistent wounds.

Vitamin D3 shows antifungal promise
Antifungal activity of vitamin D against Candida albicans in vitro and in vivo.
Most Relevant Research
We explored the antifungal effects of vitamin D3 against yeast infections, focusing on Candida albicans, in a thorough study. Using a variety of methods, including a broth microdilution test and solid plate assays, we found that vitamin D3 significantly inhibits the growth of different Candida species in a way that was both broad and dose-dependent.

Additionally, vitamin D3 showed a strong antifungal effect during the crucial phases of biofilm formation—this is important because Candida can often form protective layers that make infections harder to treat. We investigated the mechanism behind this action and discovered that vitamin D3 affects several metabolic pathways within the yeast, suggesting that it operates through multiple targets.

In an experimental model of intra-abdominal candidiasis, we noted that vitamin D3 reduced fungal presence in key organs like the liver and kidneys. Furthermore, analysis revealed that treatment with vitamin D3 resulted in less inflammation, as indicated by decreased inflammatory cell presence and lower levels of specific cytokines in the blood. Overall, these findings propose vitamin D3 as a promising new treatment option for combating yeast infections, particularly in difficult-to-treat cases like intra-abdominal candidiasis.

Vitamin D3 improves back pain
Vitamin D Supplementation in Patients with Chronic Low Back Pain: An Open Label, Single Arm Clinical Trial.
Most Relevant Research
We sought to understand the role of vitamin D3 supplementation in alleviating chronic low back pain (CLBP). In a single-arm, open-label study at a tertiary care hospital, we worked with 68 patients suffering from CLBP for at least three months, each demonstrating a significant pain level on a visual scale alongside low vitamin D levels.

Participants received 60,000 IU of vitamin D3 weekly for eight weeks. We tracked pain intensity and functional disability at various stages: at the outset, and then at two, three, and six months post-supplementation. Notably, we witnessed a remarkable increase in vitamin D levels from an average of 12.8 ng/mL to 36.07 ng/mL in those eight weeks, with 66% of participants achieving normal vitamin D levels.

The benefits didn’t stop there. Patients reported a significant drop in their pain scores at all follow-up periods, improving from 81 to 36 on the scale. Functional ability also saw enhancement over time, proving the positive influence of vitamin D3 on both pain and everyday activities.

Overall, our findings suggest that vitamin D3 supplementation can be an effective avenue for improving pain intensity and functionality in individuals with low back pain, warranting further investigation through controlled clinical trials.

Our findings revealed a significant correlation between lower vitamin D3 levels and higher edema index (EI) percentages. Patients with EI below 100% had a median serum vitamin D3 level of 65.58 ng/mL, whereas those with EI above 100% had a much lower median of 37.33 ng/mL.

We found that for every increase of 1 ng/mL in serum vitamin D3, the extent of brain edema decreased by approximately 4%. This suggests that maintaining adequate levels of vitamin D3 may serve as a protective factor against peritumoral brain edema in patients with meningioma.

Vitamin D3 reduces thyroid autoimmunity
A Prospective Study to Evaluate the Possible Role of Cholecalciferol Supplementation on Autoimmunity in Hashimoto's Thyroiditis.
Most Relevant Research
We explored the potential effects of vitamin D3, or cholecalciferol, on individuals diagnosed with Hashimoto's thyroiditis, a common form of hypothyroidism characterized by autoimmune responses that damage thyroid cells. Our study was a carefully structured clinical trial conducted over one year, which included 100 patients suffering from both Hashimoto's thyroiditis and vitamin D deficiency.

Divided into two groups, one received vitamin D3 supplements while the other was given a placebo. After eight weeks, we measured the levels of anti-thyroid peroxidase antibodies, which indicate autoimmune activity against the thyroid.

Our findings revealed a significant reduction in these antibody levels among patients who received vitamin D3, showcasing a 30.5% decrease compared to just a 16.5% decrease in the placebo group. This suggests that vitamin D3 supplementation may play a beneficial role in managing the autoimmune activity associated with hypothyroidism.

Overall, our research shows that addressing vitamin D deficiency might be an effective strategy to help manage Hashimoto's thyroiditis, which could encourage discussion about incorporating vitamin D as part of treatment plans for patients. Further research will be necessary to confirm these findings and explore the long-term benefits of vitamin D supplementation.

Vitamin D3 supplementation effective
Vitamin D in blood serum and chronic pancreatitis.
Most Relevant Research
We explored the relationship between vitamin D levels and chronic pancreatitis, a condition often characterized by malabsorption and malnutrition. In a study involving fifty patients with chronic pancreatitis and a control group of twenty individuals without gastrointestinal diseases, we measured the vitamin D levels in their blood serum.

Our findings revealed that a significant number of patients with chronic pancreatitis—about 60-80%—had notably low vitamin D levels. This is concerning, as low vitamin D can contribute to various health issues. To address this, we administered a high dose of 1.5 million IU of vitamin D3 daily to those with low levels for a duration of 24 weeks. After this treatment, we reassessed the participants' vitamin D levels.

The results were promising; vitamin D levels in patients improved significantly following supplementation. However, while we observed effective correction of vitamin D deficiency, more research is essential to determine potential long-term benefits, particularly regarding its role in fibrogenesis. Our study's limitations include the relatively small number of subjects, which suggests the need for broader studies in the future.

Vitamin D3 may prevent prostate cancer
1,25-Dihydroxyvitamin D Suppresses Prognostic Survival Biomarkers Associated with Cell Cycle and Actin Organization in a Non-Malignant African American Prostate Cell Line.
Most Relevant Research
We explored how vitamin D3, specifically the metabolite 1α,25(OH)D, influences the behavior of non-cancerous prostate cells derived from African American men. In our study, we treated the RC-77N/E cell line with vitamin D3 and observed significant changes in gene expression after just 24 hours.

Our analysis revealed that 1α,25(OH)D affected 1,601 genes, highlighting its potential to suppress pathways linked to prostate cancer progression and cell proliferation. This points to the ability of vitamin D3 to help maintain non-malignant cell characteristics.

Additionally, we identified two specific genes, Anillin and Epithelial Cell Transforming 2, which were significantly downregulated by vitamin D3 treatment and correlated with better overall survival in prostate cancer patients. This suggests that these genes could serve as valuable biomarkers for assessing the risk of disease in affected individuals.

Given these promising findings, we believe that 1α,25(OH)D not only plays a vital role in regulating prostate health but could also be used as part of a preventative strategy or as an adjunct therapy, particularly for African American men who are disproportionately affected by prostate cancer.

Vitamin D3 protects neurons from stroke
Vitamin D alleviates neuronal injury in cerebral ischemia-reperfusion via enhancing the Nrf2/HO-1 antioxidant pathway to counteract NLRP3-mediated pyroptosis.
Most Relevant Research
We explored the potential benefits of vitamin D3 (specifically, 1,25-vitamin D3) for protecting the brain during events such as strokes caused by impaired blood flow, known as cerebral ischemia-reperfusion injury (CIRI). In this study, rats received vitamin D3 supplementation for a week before undergoing a procedure that mimicked a stroke. The results were striking; those treated with vitamin D3 showed significant improvements in neurological function, with reduced areas of brain damage and more surviving neurons.

We also tested the effects of vitamin D3 on rat cortical neurons in a laboratory setting, specifically observing its influence on cell survival during oxygen-glucose deprivation and reoxygenation (a model for stroke). The vitamin D3 treatment not only improved cell viability but also appeared to reduce cell death and inflammation. Our analyses revealed that vitamin D3 boosts important protective proteins in the brain, which help combat oxidative stress and inhibit pathways linked to inflammatory cell death.

Overall, our findings strongly suggest that vitamin D3 could be a valuable resource in stroke management by protecting neurons through its antioxidant actions and reduction of harmful inflammation.

Vitamin D3 shows promise for Alzheimer's
Chronic vitamin D supplementation alleviates cognition impairment via inhibition of oxidative stress regulated by PI3K/AKT/Nrf2 in APP/PS1 transgenic mice.
Most Relevant Research
We explored the impact of vitamin D3 supplementation on cognitive decline in transgenic mice that model Alzheimer's disease. Over a 20-week treatment period, we conducted a series of tests, including a Morris Water Maze test to assess cognitive function and various analyses to gauge oxidative stress and Alzheimer-related pathology.

Our observations indicated that vitamin D3 treatment not only improved the memory performance of the mice but also positively influenced brain health. Specifically, we found that vitamin D3 led to a reduction in amyloid beta (Aβ) buildup, a hallmark of Alzheimer's, and decreased levels of proteins associated with the disease, such as APP, tau, and phosphorylated tau (p-tau).

Moreover, vitamin D3 appeared to bolster the brain's defenses against oxidative stress, enhancing key antioxidant levels while lowering harmful substances related to oxidative damage. The treatment activated important pathways involved in cellular protection, further supporting the notion that vitamin D3 may offer neuroprotective effects.

In conclusion, our study suggests that vitamin D3 could play a beneficial role in improving cognition and reducing Alzheimer-like changes in the brain. These findings invite further exploration into the potential of vitamin D3 as a supportive treatment in Alzheimer’s disease management.

Vitamin D3 improves epilepsy outcomes
Therapeutic effect of acute and chronic use of different doses of vitamin D3 on seizure responses and cognitive impairments induced by pentylenetetrazole in immature male rats.
Most Relevant Research
We aimed to understand how vitamin D3 affects seizures and cognitive problems related to epilepsy. In our study, we used sixty-six young male rats, dividing them into control and treatment groups. This allowed us to compare the outcomes of those treated with vitamin D3 against our control rats.

We observed that the rats with epilepsy showed notable issues with learning and memory, as their ability to retain information was impaired. Additionally, when exposed to pentylenetetrazole (a chemical that induces seizures), these rats experienced increased seizure duration and decreased latency before seizures. However, interestingly, when we administered vitamin D3, both acutely and chronically, these adverse effects were significantly improved.

Not only did vitamin D3 help reduce seizure severity, but it also positively influenced the levels of antioxidants and inflammatory markers in the hippocampus, a key brain area for learning and memory. Our findings suggest that vitamin D3 can indeed play a beneficial role in mitigating the cognitive and seizure-related challenges associated with epilepsy.

Calcitriol aids heart recovery
Novel Insights into the Cardioprotective Effects of Calcitriol in Myocardial Infarction.
Most Relevant Research
We examined the impact of calcitriol, the active form of vitamin D, on heart attack recovery using a mouse model of myocardial infarction (MI). In our study, we treated mice that had suffered a MI with calcitriol and observed promising results.

Our findings revealed that calcitriol helped reverse negative effects on heart function and reduced cardiac remodeling after a heart attack. It also targeted the inflammatory response typically associated with MI, improving the survival of heart muscle cells and promoting their regeneration.

We discovered that calcitriol works by enhancing the activity of the Vitamin D receptor (VDR). This process not only interferes with inflammatory signals but also leads to positive changes at the genetic level, further supporting heart health after an MI. Overall, our study provides strong evidence of calcitriol's cardioprotective properties, making it a potential treatment avenue for better outcomes following a heart attack.

Vitamin D3 in menopause management
Utilizing a Combination of Supplements Comprising Boric Acid, Magnesium, Vitamin D3, and Extra Virgin Olive Oil to Improve Metabolism in Menopausal Ovariectomized Rats.
Most Relevant Research
In this study, we explored how vitamin D3, among other supplements, affects metabolic health during menopause. The research utilized a group of ovariectomized female rats, which serve as a model for menopausal conditions in women. These rats were divided into several groups, including a control group and various groups that received different treatments, including vitamin D3.

The findings indicated that administering vitamin D3, along with boric acid, magnesium, and extra virgin olive oil, led to notable improvements in metabolic parameters. We observed benefits in areas such as inflammation and oxidative stress, which are crucial for managing health during menopause. However, while vitamin D3 showed positive effects, the study highlighted that the combination of all supplements had a more significant impact than vitamin D3 alone.

Ultimately, while vitamin D3 contributes positively to metabolic health in menopause, the full extent of its isolated benefits remains unclear without further studies. Thus, we recognize the need for additional research to fully understand how effective vitamin D3 can be independently in this context.

We focused on understanding the role of vitamin D3 in treating chronic rhinosinusitis (CRS), a condition affecting the nasal passages and sinuses. Our study analyzed 200 patients with vitamin D deficiency, dividing them into two groups: one group was given oral vitamin D supplements, while the other received a placebo.

Over a span of three months, we monitored changes in vitamin D levels and symptoms using the Total Nasal Symptom Score (TNSS). Initially, the average vitamin D level was quite low at 12.31 ng/ml. Remarkably, after treatment, this level surged to 29.71 ng/ml, indicating a significant improvement.

Additionally, we observed a reduction in TNSS scores, showing that while the average score started at 11.92, it decreased by about 10.65 points after three months of vitamin D treatment. This significant drop highlights the positive impact vitamin D supplementation can have on alleviating symptoms of CRS.

Overall, our exploration revealed a previously high prevalence of vitamin D deficiency among CRS patients, suggesting that addressing this deficiency through supplementation can be beneficial in managing their symptoms effectively.

Vitamin D deficiency linked to UTI
The Association between Vitamin D Levels and Urinary Tract Infection in Children.
Most Relevant Research
We aimed to explore the possible connection between vitamin D levels and urinary tract infections (UTIs) in children. Our study involved measuring various health markers, including vitamin D levels, in 82 children who had experienced their first UTI and compared these results to 64 healthy controls.

The findings revealed that children with UTIs had significantly lower levels of vitamin D compared to their healthy peers. Specifically, those with acute pyelonephritis exhibited even lower vitamin D levels than those with lower UTIs. We also noted that girls tended to have lower vitamin D levels than boys.

Multivariate analysis indicated that a vitamin D level below 20 ng/ml was associated with a higher likelihood of UTIs in children. Therefore, our results suggest that vitamin D deficiency may indeed pose a risk for urinary tract infections in this population.

Vitamin D3 may reduce influenza severity
Oral Supplementation of the Vitamin D Metabolite 25(OH)D Against Influenza Virus Infection in Mice.
Most Relevant Research
We aimed to understand how oral supplementation of 25-hydroxyvitamin D, a key metabolite of vitamin D3, influences influenza A virus infection in mice. In our study, mice were given a diet enriched with a high dose of 25(OH)D before they were exposed to the influenza virus.

The results were promising. Mice that received the vitamin D3 supplement showed significantly lower viral levels in their lungs compared to mice that were fed a standard diet. Furthermore, we noticed a decrease in certain pro-inflammatory cytokines, namely IL-5 and IFN-γ, which are involved in the body's inflammatory response to infection. Importantly, we found that the anti-inflammatory cytokines did not see a significant increase.

These findings suggest that 25(OH)D can help to suppress excessive inflammatory responses and may reduce both the replication of the virus and the overall severity of influenza in this mouse model. It opens the door to further exploration on vitamin D3's role in combating influenza in humans.

Vitamin D reduces triglycerides effectively
Effect of Vitamin D Supplementation on Lipid Profile in Overweight or Obese Women: A Meta-analysis and Systematic Review of Randomized Controlled Trials.
Most Relevant Research
We explored the effects of vitamin D supplementation on lipid profiles in overweight and obese women. Our investigation included data from various randomized controlled trials (RCTs) that specifically examined how vitamin D might influence cholesterol and triglyceride levels in this population.

Our analysis revealed that vitamin D had a significant impact on lowering triglycerides and total cholesterol levels. We found reductions of about 6.13 mg/dL for triglycerides and 4.45 mg/dL for total cholesterol. Additionally, there was a noteworthy increase in HDL cholesterol, the “good” cholesterol, by an average of 1.54 mg/dL.

Interestingly, we noticed that those with higher baseline triglyceride levels saw even greater benefits, particularly with a significant reduction in triglycerides when vitamin D was supplemented for 26 weeks or less. However, it’s important to mention that vitamin D supplementation did not have a significant effect on LDL cholesterol levels in this group.

Overall, our findings suggest that vitamin D can be an effective option for managing certain aspects of lipid profiles in overweight or obese women, particularly in addressing high triglycerides.

Vitamin D3 ethosomes for psoriasis
Synthesis of vitamin D3 loaded ethosomes gel to cure chronic immune-mediated inflammatory skin disease: physical characterization, in vitro and ex vivo studies.
Most Relevant Research
We aimed to explore the effectiveness of a special gel infused with vitamin D3, known as an ethosome, in managing psoriasis. This inflammatory skin condition is driven by an overactive immune response, and vitamin D3 plays a crucial role in the growth and development of skin cells called keratinocytes, making it a promising treatment option.

In our research, we created formulas containing different amounts of soya lecithin, propylene glycol, and ethanol to optimize the ethosome gel. After careful testing, we found that these ethosomes had a remarkable drug encapsulation efficiency of 96.25%. They ranged in size from about 148 to 657 nanometers, showing a stable, negatively charged profile that suggests they are safe for use.

Further analyses confirmed that the ethosome gel effectively delivered vitamin D3 through the skin and maintained its stability over six months. The results indicated that an impressive percentage of the medication penetrated the skin barrier, which is essential for its effectiveness in treating psoriasis flare-ups. Our findings suggest that the ethosome formulation enhances the therapeutic potential of vitamin D3, offering a promising avenue for treating patients with psoriasis.

Vitamin D3 reduces BPPV recurrence
Randomized Controlled Trial Assessing Vitamin D's Role in Reducing BPPV Recurrence in Older Adults.
Most Relevant Research
We investigated how supplementing vitamin D3 could affect the recurrence of Benign Paroxysmal Positional Vertigo (BPPV) in older adults. In a carefully designed double-blind clinical trial, we followed participants for 12 months to assess whether those treated with vitamin D3 would experience fewer BPPV episodes compared to those given a placebo.

Participants who were low on vitamin D3 received 13 weeks of a higher dose followed by a lower one, while others with sufficient levels were simply monitored. Our results were promising: the group receiving vitamin D3 experienced an impressive 87% reduction in recurrence rates and a significant delay in the time to first recurrence compared to the placebo group.

However, we didn't find any clear differences in outcomes between those treated with vitamin D3 and the participants who were already vitamin D replete. This trial provides a strong basis for further exploring vitamin D as a standard treatment option for BPPV in future research phases.

Vitamin D3 may aid inflammation
Elevated Pro-inflammatory Cytokine Levels in Acute Reflux Esophagitis Are Reduced by 1,25 Dihydroxy Vitamin D3.
Most Relevant Research
We aimed to understand whether vitamin D3 can have a beneficial effect on reflux esophagitis (RE), a condition caused by stomach acid irritating the esophagus. To explore this, we conducted experiments on seventy male Sprague-Dawley rats, dividing them into groups to study both acute and chronic forms of RE. We treated some of these rats with varying doses of vitamin D3, while others did not receive this treatment, allowing us to compare differences in inflammation levels.

The results showed that the levels of important pro-inflammatory proteins—IL-1β, IL-6, and IL-8—were significantly elevated in the rats suffering from RE. However, when we treated the rats with vitamin D3, we observed a notable reduction in these inflammatory markers, but only in the acute RE group. Unfortunately, similar benefits were not evident in the chronic RE group, and vitamin D3 did not improve the overall condition of the esophagus on a structural level.

Interestingly, our histopathological examinations revealed varying degrees of esophageal damage, regardless of vitamin D3 treatment. While elevated serum calcium levels were noted following vitamin D3 treatment, this did not translate into improvements in chronic RE cases. Overall, our findings point to a potential protective role of vitamin D3 in early-stage reflux esophagitis, suggesting that it might help manage inflammation at the beginning of this condition.

Vitamin D3 aids liver health
Effects of vitamin D supplementation on the glycaemic indices, lipid profile and liver function tests in patients with cirrhosis: a double-blind randomised controlled trial.
Most Relevant Research
We aimed to explore how vitamin D3 supplementation affects liver disease, specifically in patients with cirrhosis. In a thorough double-blind, randomized controlled trial involving 60 participants, we offered patients either a weekly dose of 50,000 IU of vitamin D or a placebo for 12 weeks. Our goal was to assess various health indicators, including serum vitamin D levels, blood glucose measures, and liver function tests.

After the 12-week period, we observed significant changes among those taking vitamin D. The serum levels of 25-hydroxy-vitamin-D3 increased substantially, alongside notable improvements in fasting blood glucose and insulin resistance, measured by HOMA-IR. These results suggest that vitamin D3 could play a beneficial role in managing certain metabolic aspects of liver disease in cirrhosis patients.

Overall, our study highlights the potential of vitamin D3 supplementation as a simple yet effective strategy to improve specific health parameters in people living with cirrhosis, paving the way for further exploration in this important area of research.

Vitamin D may influence blood clotting
Relationship between vitamin D deficiency and thrombus load in patients with ST-elevation myocardial infarction.
Most Relevant Research
We explored the connection between vitamin D levels and coronary thrombus load in patients who experienced ST-elevation myocardial infarction (STEMI). The study included 77 patients where we measured serum vitamin D levels, thrombus degree, and other relevant factors.

Our findings revealed that a concerning 79.22% of participants showed vitamin D deficiency, with those having milder thrombus loads possessing significantly higher vitamin D levels compared to those with severe thrombus loads. Moreover, as we assessed the severity of coronary artery condition, we noted that patients with higher thrombus burden also had markedly greater scores of atherosclerosis.

Importantly, we found clear negative correlations between vitamin D levels and measures of thrombus burden. Lower vitamin D levels aligned with higher TIMI frame counts—meaning more extensive vascular obstruction—suggesting that vitamin D might play a role in blood clot formation.

Overall, we determined that low 25(OH)D3 levels could be an independent predictor of high coronary artery thrombus load in STEMI patients. These findings highlight the potential significance of monitoring and perhaps addressing vitamin D levels in this vulnerable group.

Vitamin D treatment reduces BPPV
Reduction of recurrence rate of benign paroxysmal positional vertigo by treatment of severe vitamin D deficiency.
Most Relevant Research
We sought to understand the impact of severe vitamin D deficiency on benign paroxysmal positional vertigo (BPPV), a common cause of dizziness. Previous studies showed a connection between low vitamin D levels and both the onset and recurrence of BPPV, but none had conclusively demonstrated that addressing this deficiency could lessen the recurrence of symptoms.

To examine this further, we included patients diagnosed with unilateral, idiopathic posterior canal BPPV and severe vitamin D deficiency, indicated by levels of 25-hydroxyvitamin D3 at or below 10 ng/ml. After thorough evaluations including hearing tests and balance assessments, we prescribed vitamin D therapy to our participants.

Over the following months, we monitored the changes in their vitamin D levels and followed up with them for 18 months to observe any recurrence of dizziness. Our findings revealed that patients who showed an improvement in their vitamin D levels experienced a significant reduction in the recurrence of BPPV symptoms.

This suggests that by addressing vitamin D deficiency, we can potentially offer a more effective approach to managing BPPV—lessening those troublesome dizzy spells that can really impact quality of life.

Vitamin D3 improves migraine symptoms
Vitamin D3 might improve headache characteristics and protect against inflammation in migraine: a randomized clinical trial.
Most Relevant Research
We explored the potential benefits of vitamin D3 supplementation for people suffering from migraines. In a carefully designed study, 80 individuals with episodic migraines participated in a 12-week trial where they were randomly assigned to receive either 2000 IU (50 μg) of vitamin D3 daily or a placebo.

Throughout the study, we closely monitored headache characteristics using diaries and measured various serum inflammatory markers before and after the trial. Our findings showed that those taking vitamin D3 experienced significant improvements. They reported fewer headache days each month, shorter headache durations, and less severe pain overall when compared to the placebo group.

Interestingly, while the levels of some inflammatory markers didn't change much, we did notice a striking reduction in a certain enzyme linked to inflammation in the vitamin D3 group. Additionally, there was a trend toward lower levels of another inflammation marker, suggesting that vitamin D3 might offer a protective effect against neuro-inflammation in migraine sufferers.

In essence, our study indicates that vitamin D3 could be a promising supplemental treatment to help improve the quality of life for those battling episodic migraines.

Vitamin D3 does not prevent stones
Effect of two vitamin D repletion protocols on 24-h urine calcium in patients with recurrent calcium kidney stones and vitamin D deficiency: a randomized clinical trial.
Most Relevant Research
We assessed how two different vitamin D3 repletion protocols affect patients with recurrent calcium kidney stones and vitamin D deficiency. This was a controlled clinical trial involving 62 participants who had low levels of vitamin D and were prone to kidney stones. We administered either 2000 IU of cholecalciferol daily for 12 weeks or 50,000 IU weekly for 8 weeks.

Both treatment approaches resulted in a significant increase in 24-hour urine calcium levels; however, there was no difference between the two methods. Importantly, neither regimen increased the supersaturation of calcium oxalate or calcium phosphate, which are substances that can contribute to the formation of kidney stones.

We also observed a notable increase in serum levels of 25-hydroxyvitamin D, particularly in the weekly high-dose group, and a decline in serum parathyroid hormone in both groups. Despite these changes, it's crucial to highlight that our findings indicate no significant advantages in preventing kidney stones from vitamin D3 treatment alone.

No significant benefit from vitamin D3
Vitamin D₃supplementation and childhood diarrhea: a randomized controlled trial.
Most Relevant Research
We conducted a thorough investigation into how vitamin D3 supplementation affects diarrhea in young children. The study involved 3,046 infants aged 1 to 11 months living in high-risk areas of Kabul, Afghanistan. They were randomly assigned to receive either six doses of vitamin D3 or a placebo over 18 months.

Our analysis measured instances of diarrhea, defined as having three or more loose stools in a single day. We focused on comparing the time to the first diarrhea episode and the risk of recurring episodes between both groups.

However, the results were quite clear: vitamin D3 supplementation did not lead to any significant reduction in the time before the first episode of diarrhea or in the likelihood of having recurrent diarrhea. The differences observed between the vitamin D3 group and the placebo group were not meaningful.

Despite our expectations, this study suggests that vitamin D3 supplementation doesn't provide the preventive benefits we were hoping for when it comes to diarrhea in this population. We recommend against using similar supplementation under comparable circumstances without further research.

Additional studies in other settings might help in understanding if vitamin D3 could play a beneficial role in preventing diarrhea elsewhere.

Vitamin D3 improves adolescent insomnia
High-dose Vitamin D Supplementation and Improvement in Cognitive Abilities, Insomnia, and Daytime Sleepiness in Adolescent Girls.
Most Relevant Research
We studied how high-dose vitamin D3 supplementation might affect insomnia in adolescent girls. Over nine weeks, 940 participants received a weekly dose of 50,000 IU of vitamin D3 while we tracked changes in their sleep patterns.

Interestingly, we observed a notable reduction in insomnia rates after the treatment. The number of girls reporting insomnia dropped from 15.0% to 11.3%, indicating that this vitamin can potentially improve sleep quality.

We also noticed a slight decrease in daytime sleepiness, with cases falling from 15.6% to 14.7%. Furthermore, those experiencing both insomnia and sleepiness saw a decline from 8.0% to 6.1%.

These findings suggest that high-dose vitamin D can play a significant role in alleviating sleep issues among adolescent girls. However, further studies are needed to see if these effects are consistent across diverse age and gender groups.

Vitamin D3 improves mild sleep apnea
The effects of vitamin D supplemantation on prognosis in patients with mild obstructive sleep apnea syndrome.
Most Relevant Research
We set out to evaluate what effect vitamin D3 supplementation might have on patients suffering from mild obstructive sleep apnea syndrome (OSAS). In this study, 19 adult men who were diagnosed with mild OSAS and had low vitamin D levels participated over an 8-week period. Each week, they took a sizable dose of 50,000 IU of vitamin D3 while undergoing various health assessments. We measured important factors like blood glucose, lipid profiles, and inflammation markers, along with repeated sleep monitoring through polysomnography.

By the end of the study, we observed a significant increase in participants' vitamin D levels, which jumped from an average of 19.5 to 41.8 ng/mL—an indication that the supplementation was effective. Along with this rise in vitamin D, we also noted improvements in fasting blood glucose and total cholesterol levels, as well as a decrease in key inflammation markers. Most encouragingly, the frequency of obstructive apneas and other related sleep disturbances significantly decreased.

Based on these findings, it seems that vitamin D supplementation can play a beneficial role in improving the prognosis for those with mild OSAS. It's exciting to consider how something as simple as a vitamin could lead to better sleep health for individuals struggling with sleep apnea.

Vitamin D3's impact on constipation inconclusive
Effectiveness of the Combination of Probiotic Supplementation on Motor Symptoms and Constipation in Parkinson's Disease.
Most Relevant Research
We explored the effects of probiotic supplementation combined with vitamin D3 on constipation and motor symptoms in individuals with Parkinson's disease. This study focused on a group of patients who had chronic constipation and used a specific probiotic formula.

Over the course of three months, we evaluated 32 patients to see if the supplementation made a difference in their digestive health and overall symptoms. We carefully monitored factors like bowel habits and employed various assessments to track the subjects' progress.

While our findings indicated some benefits from the probiotic intervention, the specific impact of vitamin D3 alone on constipation remains unclear due to the combination with probiotics in our approach. Therefore, we cannot draw definitive conclusions about the effectiveness of vitamin D3 by itself in alleviating constipation. Our research does suggest that the gut microbiome plays a role in managing symptoms associated with Parkinson's disease, emphasizing the importance of ongoing investigations in this field.
